چکیده

In a great many cases the participation of singlet oxygen in sensitized photooxigenation processes in solution has been stated Singlet oxygen has sometimes also been assumed to be the important intermediate in photodynamic reactions2. Unfortunatly in most instances a direct proof by spectroscopic methods is practically impossible because of its low emission quantum yield in solution. Therefore evidence has frequently only been produced by more or less specific chemical reactions and by the interpretation of kinetical data, especially of quenching reactions 3.

Mechanisms of photodynamic inactivation of acridine orange-sensitized transfer RNA: participation of singlet oxygen and base damage leading to inactivation.

Photodynamic action/Acridine orange/tRNA/Guanine/Singlet oxygen The photodynamic inactivation of acridine orange (AO)-sensitized tRNA was investigated with respect to the type of photochemical pathways and the base damage leading to the inactivation. The diagnostic tests for singlet oxygen (1O2), namely the enhancement effect of deuterium oxide (D2O) and the inhibition effect by sodium azide, i...

On the mechanism of the acridine orange sensitized photodynamic inactivation of lysozyme. I. Basic kinetics.

The kinetics of the photodynamic desactivation of lysozyme in presence of acridine orange as the sensitizer have been investigated in detail varying oxygen, protein, dye concentration, ionic strength and pH value. The kinetics can be approximately described as an over all pseudo-first-order rate process. Changing the solvent from water to D2O or by quenching experiments in presence of azide ion...

On the mechanism of the acridine orange sensitized photodynamic inactivation of lysozyme. II. Kinetics in presence of N-acetylglucosamine.

The photodynamic deactivation of lysozyme in presence of acridine orange is caused by a reaction between singlet oxygen formed via the dye triplet state and the protein. In order to identify the region where the singlet oxygen reacts with the protein we have investigated the kinetics of the deactivation in presence ofthe inhibitor of the enzymatic reaction N-acetylglucosamine (GlcNAc). The over...

Tryptophan-dependent sensitized photoinactivation of colicin E1 channels in bilayer lipid membranes.

The bacterial toxin colicin E1 is known to induce voltage-gated currents across a planar bilayer lipid membrane. In the present study, it is shown that the colicin-induced current decreased substantially upon illumination of the membrane in the presence of the photosensitizer, aluminum phthalocyanine. This effect was almost completely abolished by the singlet oxygen quencher, sodium azide. Usin...

Esr Triplet Exciton Spectrum of Acridine Orange

The triplet state of acridine orange dissolved in methanol/water matrix was investigated by ESR. In absence of oxygen a strong temperature dependence of the spectra was observed. At low temper ature (100 °K) the zero-field splitting parameters calculated from the triplet spectrum are: X /h c = 0.0050 cm-1 , Y / h c = 0.0342 cm-1 , Z //ic = 0.0387 cm-1 , at higher temperature (140 °K) : X * ( h...
